An impressive bonus point victory at Murrayfield helped Edinburgh overtake Glasgow and go second in the Magners League standings.
Mike Blair's try and Phil Godman's accurate kicking gave the home side a 16-6 half-time advantage, Dan Parks scoring two penalties for Glasgow.
Simon Webster crossed early in the second half to extend Edinburgh's lead and Godman took his points tally to 24.
Hugo Southwell and Ben Cairns added further tries for Andy Robinson's side.
The result means Edinburgh hold a first-leg advantage in their quest to win the 1872 Challenge Cup, with the Scottish sides set to meet again at Firhill early in the New Year.
Godman, who was assuming kicking duties in the absence of Chris Paterson, put the home side into an early lead with a penalty.
Glasgow stand-off Dan Parks missed with his first penalty attempt shortly before making amends with a confident kick to level.
After another Parks miss, Warriors hooker Dougie Hall was penalised for binding and Godman regained Edinburgh's three-point lead with a kick from distance.
Edinburgh captain Blair got the game's opening try after intercepting a loose Parks pass and powering forward to cross the line. Godman added the conversion.
Warriors were caught offside after Craig Hamilton and Cairns burst through the visiting defence and Godman again showed his prowess from range to put Edinburgh further ahead.

John Barclay almost slipped through for Glasgow's first try after a line-out went astray but the flanker was penalised for knocking the ball on.
Parks reduced Edinburgh's lead with his second successful penalty of the game before Scott Newlands was sin-binned for the home side for an infringement.
Kelly Brown thought he had reduced Glasgow's deficit early in the second half when he chased Parks' up-and-under with Edinburgh's John Houston and forced the ball down beyond the try-line.
But, after consulting the TMO, referee Neil Paterson disallowed the try.
Edinburgh got their second try of the match when Godman's clever reverse pass allowed wing Webster to burst through and touch down. Godman added the conversion.
Two more Godman penalties followed as Glasgow struggled to cope with the pace and power of Edinburgh's attack.
An Edinburgh line-out led to Southwell's try as the home side's cross-field movement created space for the full-back on the left-hand side.
The Scottish capital side continued to press forward but frantic Glasgow tackling prevented Southwell registering his second try.
However, Southwell then unlocked the Glasgow defence with a pass to Cairns, who went on to score Edinburgh's final try, with replacement David Blair failing to convert.
